Festival organiser John Giddings said he has wanted the Strokes to appear at the festival every since he heard their first album, adding that he cannot wait until the event next year.
Friday will see Jay-Z take to the stage while the Strokes will appear on the Saturday of the ninth Isle of Wight festival.
Orbital will also appear on the Saturday, headlining the Big Top, while Squeeze will perform an exclusive gig for campers on the Thursday night.
"Jay-Z and Pink are two of the best live acts I have ever seen and the Blondie hits speak for themselves. I can't wait," added Mr Giddings.
The Big Top will be back this year, featuring Alesha Dixon, Calvin Harris, McFly, Noisettes, Pixie Lott and The Charlatans.
Tickets go on sale on December 4th for the event on June 11th to 13th 2010.
